Sleep that broke, and the number that came from eight men.

Three a.m. is a whole personality in perimenopause. So the 'sleep stack' sells itself. I wanted to know where the numbers in those protocols came from. It was a short trip.

The short version

DSIP was the one peptide the FDA advisory committee voted against in July 2026. Ipamorelin's only human pharmacology comes from eight healthy men per dose level. No human trial of CJC-1295 with ipamorelin for sleep exists, in either sex. And the molecule in most 'CJC' vials isn't the one that was studied.

7–6–1the FDA committee vote against DSIP, July 2026.

What gets sold for it

The standard 'sleep stack' is CJC-1295 with ipamorelin, sometimes with DSIP (delta sleep-inducing peptide, which is a great name), sometimes with a growth-hormone secretagogue like sermorelin. The story is that growth hormone pulses during deep sleep, so nudging growth hormone nudges deep sleep. Neat. Untested.

What's actually been studied

DSIP. In July 2026 the FDA's advisory committee reviewed seven peptides for the compounding list. Six got a favourable (advisory, non-binding) vote. DSIP was the one rejected, 7–6–1. If you see it on a menu this month, ask the person selling it whether they know that.

Ipamorelin. The only human pharmacokinetic study is from 1999 and used eight healthy male subjects per dose level. Every half-life, clearance figure and dose number in circulation comes from those men. Two later Phase 2 trials in post-surgical patients finished in 2009 and 2014 and never posted results, which in drug development usually means what you think it means. There is no human trial of ipamorelin for sleep, body composition or anything else you'd buy it for, in either sex.

CJC-1295. Two red flags nobody discloses. One: the single human study tested CJC-1295 with DAC, a version with a half-life measured in days. Most of what's sold is CJC-1295 without DAC, half-life about thirty minutes, with no published human trials at all. You often can't tell which you've got. Two: the only registered efficacy trial CJC-1295 ever had was terminated in 2006 with no reason posted and was never published.

Sermorelin. It was an approved drug once — as a diagnostic agent and for pediatric growth-hormone deficiency. It was withdrawn commercially in 2008. 'FDA-approved' on a sleep menu is doing a lot of work in that sentence.

The part that's specific to us

Perimenopausal sleep breaks for hormonal reasons — progesterone drops, night sweats, the 3 a.m. cortisol bump — and none of the compounds above were tested against any of it. The women on the perimenopause forums who say their HRT did more for sleep than anything in a vial are not wrong to say so. Progesterone, in particular, has actual sleep data.

And if you're on oral estrogen, there's a mechanism by which the whole GH-secretagogue category works against itself. It's on the doctor-questions page.

What we order

Honestly? Less than you'd think, for this one. The women I know who research sleep compounds order CJC/ipamorelin from ZestyRat, look at the batch COA, and go in with their eyes open about the evidence above. Nobody I know is ordering DSIP since the vote. The supplier is the one variable in this we can actually control, so we control it.

Questions I actually get

Which sleep peptide actually works?

No compound sold as a 'sleep peptide' has a human sleep trial behind it. That's the finding. If a page tells you otherwise, ask for the study.

Is sermorelin FDA-approved?

It was, for a diagnostic use and for children with growth-hormone deficiency, and the approval was withdrawn for commercial reasons in 2009. It can be lawfully compounded for that reason. That is a legal fact, not an evidence-for-sleep fact.

Where do you order?

ZestyRat, code SHAUN10 — and I'd read the section above twice before ordering anything for this.

The runner-up, if you want the pretty one.

Amino Club is the one that looks like it was designed for us — soft branding, ISO 17025 third-party COAs, US shipping. It's the group chat's second vote, and the code SHAUN22 works there. Why it's number two, not number one →

Nothing on this page is medical advice or a recommendation to use anything. The compounds discussed are sold as research materials and are not FDA-approved for the uses people talk about. I read; I don't prescribe. Talk to a clinician who knows your history.

One honest letter a month

No hype. No protocol. No selling you a plan.

What I'm reading, what changed price, what the group chat is talking about — and the things that turned out to be nothing.
